				Bacterial Expression Systems
Broad range of bacterial expression systems

		1) E. COLI EXPRESSION SYSTEMS
The pET Expression System
The pET expression system is offered with of N- or C-terminal 6xHN-tagged vectors together with IMAC-based purification. Two levels of regulation provide the highest level of protein expression. First level is ensured by pET6xHN series of vectors, second level is secured in host cells.
Main features:
- Higher inducible protein expression levels and tighter control of your target gene
- E. coli BL21(DE3)-based system
- Choose between N- & C-terminal 6xHN-tagged vectors
- Choice of fast, simple In-Fusion PCR cloning or traditional T4 DNA ligase cloning
- Purify your expressed protein with His60 Nickel resin or TALON Cobalt resin
Single Protein Production (SPP) System
This modified version of cold-shock expression vectors along with expression of the endoribonuclease MazF ensures protein purity and stable isotopic labeling protein labeling. Target protein production starts 72 hours after induction.
Main features:
- Convenient for radioisotope labeling
- Up to 90% of newly expressed cellular protein is labeled target protein
- Suitable for NMR analysis
- Purified protein expression
HAT-tag expression system
This system utilizes HAT (histidine affinity tag) in protein expression and specifically protein purification. Novel polyhistidine epitope tag encoded in HAT vector allows purification of protein in neutral or physiological pH level or native conditions.
Main features:
- Purify proteins at neutral pH
- Use under native or denaturing conditions
- Higher protein solubility
- Obtain optimum purification with TALON IMAC resin
2) BREVIBACILLUS EXPRESSION SYSTEMS
Brevibacillus Expression System
System is suitable for eukaryotic recombinant protein expression with high yield of active protein. Products are kept intact thanks to system being protease free. His-tag vectors (pNC-HisE, pNC-HisF, pNC-HisT, pNI-His) are also available for quick and easy purification of the expressed target proteins.
Main features:
- Production of secreted or intracellular recombinant target proteins
- Products remain intact in culture medium
- Unlike E. coli, no endotoxins
- Proteins are produced in active form
- Easy to culture, handle and sterilize
3) B. SUBTILIS EXPRESSION SYSTEMS
Bacillus subtilis Secretory Protein Expression System
Bacillus subtilis has the ability to secret protein directly into the culture media, amenability to medium- and large-scale fermentation and lack codon bias. Secretory Protein Expression System from Takara Bio allows rapid development of a B. subtilis clone library, with each clone bearing a pBE-S construct in which the ORF of your protein of interest is fused with 173 different sequences for unique signal peptides.
Main features:
- Includes pBE-S DNA, an E. coli/B. subtilis shuttle vector containing:
- B. subtilis-derived subtilisin (aprE) promoter
- Secretory signal peptide (aprE SP)
- Multiple cloning site (MCS)
- 3' (C-terminal) His-tag sequence

- Supplied with SP DNA Mixture, a library of DNA sequences encoding 173 unique secretory signal peptides
- Fully compatible with In-Fusion cloning kits and systems, allowing rapid and easy construct generation
- Includes B. subtilis strain RIK1285
