Cre Recombinase

The unique and specific recombination system allows to edit genes in application as gene knockins or knockouts

Cre recombinase is enzyme with the site-specific recombinase function, which mediates recombination between the two 34 base pairs loxP sites. It can be used to to carry out deletions, insertions, translocations, and inversions. Takara oferrs two effective ways of Cre recombinase delivery – gesicles and AAV. Cre RLPs are also to be found in the portfolio.

1) GESICLES

Delivery of Cre recombinase protein without need of nucleic acid presence usin nanovesicles. Whole proces sis much easier then using plasmid or viral delivery systems. Cre recombinase is present in active state, so no delay for transcription and translations occures.

Main features:

  • For knockin and knockout studies
  • No preparation or pre-treatment of target cells required
  • Works even with dividing and non-dividing cells, primary cells, and cell lines
  • No risk of Cre recombinase expression in the target cells 

2) AAV PARTICLES

AAVpro® Helper Free System (AAV2-CRE Recombinase) is designed for preparation of serotype 2 AAV viral particles to deliver the Cre recombinase into the mammalian cells. Kit generates high titer without need of helper virus.

Main features:

  • Delivery if Cre recombinase gene to hard-to-transfect mammalian cells
  • Suitable also for proliferating and non-proliferating cells
  • Includes an AAV Extraction Solution for efficient virus isolation

3) Cre RLPs

Cre RNA lentiviral particles (RLPs) enables delivery of active Cre mRNA into mammalian cells using nonintegrating, virus-like particles (RLPs). No viral genom is present or generated during the process. RLPs proved high efficiency and mediate transient expression in in vitro and in vivo applications involving cell lines, primary cells, stem cells, tissues, and organisms.

Main features:

  • Can be used with immortalized cell lines, dividing and nondividing primary cells, stem cells, tissues, and various model organisms
  • No viral genome and risks associated with random genomic integration
  • Available as biologically active Cre recombinase and ZsGreen1 mRNAs packaged in RLPs
