Immune Profiling

Takara Bio kits combine SMART (Switching Mechanism at 5' End of RNA Template) technology with 5' RACE to capture V(D)J sequences. With RNA as input material kits detect expressed receptor sequences of B-cell and T-cell receptors. Sequencing libraries can be multiplexed with up to 96 primer index options. Separate portfolio for human and mice is available.

1. HUMAN IMMUNE PROFILING KITS

SMART-Seq Human BCR (with UMIs)

• Input: down to 10 ng of high-integrity RNA (may differ depending on starting material)
• Sample material: PBMCs, B cells, spleen or bone marrow, and whole blood
• Enables profiling of all heavy (IgG, IgM, IgA, IgD, IgE) and light (IgK, IgL) BCR chains
• Can be miniaturized to reduce costs, or automated

SMARTer® Human BCR IgG IgM H/K/L Profiling Kit

• Total RNA inputs: 10ng - 1 µg of RNA from blood or 1- 100 ng of RNA from B-cells
• Accurate amplification of human IgG subclasses and identification
• Enables flexible pooling strategies

SMART-Seq® Human TCR (with UMIs)

• Input: 10 ng to 500 ng of total RNA or 1,000 to 10,000 purified T cells
• Sample material: total RNA or 1000 -10,000 purified T-cells
• Single primer pair for each TCR (alpha or beta) subunit per reaction
• UMIs and UDIs implemented in the kit

SMARTer® Human scTCR a/b Profiling Kit

• Starting material: FACS or manually sorted single cells
• 96 cells can be pooled into 12 libraries
• Detection of low-abundance TCR variants

2. MOUSE IMMUNE PROFILING KITS

SMART-Seq® Mouse BCR (with UMIs)

• Input: 10 ng–1 μg of total RNA (RIN>7)
• Sample material: mouse spleen, bone marrow, or peripheral blood mononuclear cells (PBMCs)
• For includes all heavy (IgG, IgM, IgA, IgD, IgE) and light (IgK, IgL) BCR chains
• UMIs and UDIs re incorporated in the kit

SMARTer® Mouse BCR IgG H/K/L Profiling Kit

• Input: 10 ng–3 µg of total RNA
• Sample material: spleen, lymph node, PBMCs, and hybridomas
• Generates data for both heavy (IgG only) and light chain diversity
• Flexible pooling possible

SMART-Seq® Mouse TCR (with UMIs)

• 1 ng–1 µg of total RNA or 100–10,000 purified mouse T cells
• Sample material: mouse spleen or T cells, mouse whole blood, bone marrow, or thymus
• Either full-length V(D)J or CDR3 only can be sequenced
• UMIs and UDIs re incorporated in the kit
